RESUME

LATAR BELAKANG           :

Elektroforesis menerapkan prinsip pemisahan molekul bermuatan dalam suatu medan listrik. Jika ditempatkan dalam medan listrik, muatan negatif dan positif suatu biomolekul bergerak menuju elektroda yang berlawanan karena adanya gaya tarik-menarik elektrostatik. Elektroforesis bergantung pada persamaan v = E.q / f, dengan E adalah kekuatan medan listrik, q muatan total pada molekul dan f adalah koefisien gesekan. Menurut persamaan ini, molekul akan  bergerak lebih cepat apabila muatannnya diperbesar, muatan listrik diperkuat dan f menurun. Molekul muatan yang sama terpisahkan karena perbedaan koefisien gesek sementara molekul  dengan massa/bentuk yang sama mungkin sangat berbeda satu sama lain dalam hal muatannya. Sehingga campuran yang kompleks mungkin dipisahkan mencapai resolusi yang tinggi dengan proses elektroforesis.

MANFAAT                            :

Gel agarosa teknik elektroforesis banyak digunakan untuk menyelidiki efisiensi pembelahan molekul kecil dan mempelajari pola pembelahan DNA. Teknik dengan menggunakan gel agarosa ini dapat digunakan untuk mendesain sebuah obat. Pembelahan DNA yang terjadi juga digunakan untuk menghitung laju hidrolisis.

METODE                               :

Metode elektroforesis ini menggunakan gel karena gel secara kimiawi lebih tidak aktif, mereka sedikit berinteraksi dengan biomolekul selama elektroforesis sehingga memungkinkan pemisahan berdasarkan fisik daripada perbedaan kimia antara komponen sampel. Material yang sering digunakan dalam pemisahan asam nukleat dan protein adalah gel agarosa dan gel akrilamida. Gel agarosa adalah gel yang paling umum digunakan saat ini. Agarosa menghasilkan derajat yang lebih seragam daripada pati dan dapat bervariasi dengan mengubah konsentrasi mulai dari suspensi(konsentrasi rendah mengubah pori-pori menjadi besar, sementara konsentrasi tinggi mengubah pori-pori menjadi kecil). Karena distribusi muatan seragam dalam asam nukleat, untuk menentukan massa molekul DNA cukup akurat berdasarkan mobilitas pada gel agarose. Akrilamida adalah sebuah gel yang lebih tepat untuk pemisahan elektroforesis dari kedua protein dan asam nukleat. Untuk pemisahan protein, rasio akrilamida: N, metilen bis akrilamida N ' biasanya 40:1 sedangkan untuk pemisahan DNA adalah 19:01. Gel tersebut cocok untuk pemisahan DNA resolusi tinggi dan protein. Alat-alat yang digunakan adalah electrophoresis chamber(kotak elektroforesis), power supply, gel pengecoran nampan yang tersedia dalam berbagai ukuran dan terdiri dari plastik UV transparan, sisir untuk membentuk sumur-sumur sampel pada gel, buffer elektroforesis(penyangga) seperti Tris-acetate-EDTA (TAE) atau Tris-borate-EDTA (TBE), Transilluminator(kotak sinar ultraviolet), yang digunakan untuk memvisualisasikan DNA bernoda pada gel. CATATAN: selalu memakai kacamata pelindung ketika mengamati DNA pada Transilluminator untuk mencegah kerusakan mata dari sinar UV.

CARA KERJA                       :          

1. Mempersiapkan gel. Agarosa bubuk dicampur dengan buffer elektroforesis sesuai konsentrasi yang diinginkan kemudian dipanaskan dalam oven untuk mencairkannya. Etidium bromida ditambahkan ke gel (konsentrasi akhir 0,5 ug / ml) untuk memfasilitasi visualisasi DNA setelah elektroforesis.

2. Setelah larutan didinginkan  sampai dengan 60°C, larutan dituangkan ke nampan pengecoran yang mengandung sisir sampel dan didiamkan pada suhu kamar.

3. Setelah gel telah dipadatkan, sisir diambil dengan hati-hati jangan sampai merobek bagian bawah sumur.

4. Gel yang masih berada dalam plastic tray, dimasukkan secara horizontal ke kotak elektroforesis dan ditutupi dengan buffer.

5. Sampel yang mengandung DNA dicampur dengan loading buffer kemudian dipipet ke dalam sumur sampel, penutup dan power supply ditempatkan pada alat.

6. Arus diberikan.

7. Aliran arus dikonfirmasi dengan mengamati gelembung dari elektroda. DNA akan bermigrasi ke arah elektroda positif, yang biasanya berwarna merah.

8. DNA yang telah bermigrasi dalam gel dapat pantau dengan visual sesuai pewarnaan jejak DNA oleh biru bromofenol dan pewarna cyanol xylene.

9. Konsentrasi gel agarosa yang berbeda, dapat mengatasi berbagai ukuran fragmen DNA.

Finally, I'm back from a looooong journey!

Q: Where did I go?
A: Of course, I've gone to my second hometown, JAKARTA!!! Due to some bussiness, I'm forced to go there with my fam (Mom, Dad, and Rama) by CAR! Tiring, isn't it? But, enjoyment relieved the stress. I went to some places to accompany my dad meeting his relatives and I also had a chance meeting mine. I'm soooo happy :):) While, rama&mom went to Dufan Amusement Park with her colleagues.

Q: What did I do in spending my time there?
A: Most, I spent my time doin' the same thing all over again. Gone somewhere. Except for mom, she already had gone to Surabaya by plane leaving three of us behind (That is so not fair, Mom :p)

I want to write more, the complete story, but it is too much to write. So, this is enough (I think). Is it alright? (I hope :))

Park Shi Hoo❤

Have you guys known about the 2010 korean drama Prosecutor Princess?? Well it is now being aired in Indosiar everyday at 12 noon. I reaaaaaaaaaaaaally love this drama right after City Hunter❤.

This Prosecutor Princess tells about a talented young woman, Ma Hye Ri who works as a prosecutor. She really does well solving cases that happened after some humiliations (for more info please watch the drama :), sorry for not telling you guys but it will be too long for me writing the whole story). Futhermore, a former lawyer, Seo In Woo (kyaaaa he's Park Shi Hoo❤ I wish he could be mt boyf awawaw he's too good to be true~ ㅋㅋㅋ) always helps her when she needs someone to rely on. He's just toooo kind and sincere but actually behind his sincerity, there are reasons why he came to Ma Hye Ri at the very first time........ You should watch this drama, HIGHLY RECOMMENDED. Aw I forget to tell you who Park Shi Hoo is...... Now, it's almost dawn and I should've gone to sleep. So, maybe I'll tell you tumoloooow. I'm just going to post a photo. Is it alright??

Resistor adalah komponen elektronik dua saluran yang didesain untuk menahan arus listrik dengan memproduksi penurunan tegangan diantara kedua salurannya sesuai dengan arus yang mengalirinya, berdasarkan hukum Ohm:

Potensiometer


Potensiometer adalah resistor tiga terminal dengan sambungan geser yang membentuk pembagi tegangan dapat disetel. Jika hanya dua terminal yang digunakan (salah satu terminal tetap dan terminal geser), potensiometer berperan sebagai resistor variabel atau Rheostat.

Potensiometer biasanya digunakan untuk mengendalikan peranti elektronik seperti pengendali suara pada penguat. Potensiometer yang dioperasikan oleh suatu mekanisme dapat digunakan sebagai transduser, misalnya sebagai sensor joystick.


Dioda



Dioda adalah komponen aktif bersaluran dua (dioda termionik mungkin memiliki saluran ketiga sebagai pemanas). Dioda mempunyai dua elektroda aktif dimana isyarat listrik dapat mengalir, dan kebanyakan dioda digunakan karena karakteristik satu arah yang dimilikinya. Dioda varikap (VARIable CAPacitor/kondensator variabel) digunakan sebagai kondensator terkendali tegangan.


Fotodioda

Fotodioda adalah jenis dioda yang berfungsi mendeteksi cahaya. Fotodioda merupakan sensor cahaya semikonduktor yang dapat mengubah besaran cahaya menjadi besaran listrik. Fotodioda merupakan sebuah dioda dengan sambungan pn yang dipengaruhi cahaya dalam kerjanya. Cahaya yang dapat dideteksi oleh fotodioda ini mulai dari cahaya infra merah, cahaya tampak, ultra ungu sampai dengan sinar-X. Aplikasi fotodioda mulai dari penghitung kendaraan di jalan umum secara otomatis, pengukur cahaya pada kamera serta beberapa peralatan di bidang medis.


Konsep Dasar Robot

Robot berasal dari bahasa Czech, robota, yang berarti pekerja. Pada dasarnya robot dibuat untuk mendukung dan membantu pekerjaan manusia, seperti yang banyak terlihat dibidang industri dimana robot dapat meningkatkan hasil produksi industri tersebut. Robot diklasifikasikan ke dalam 4 bagian, yaitu :

 Non mobile robot

Robot ini tidak dapat berpindah posisi dari satu tempat ke tempat lainnya, sehingga robot tersebut hanya dapat menggerakkan beberapa bagian dari tubuhnya dengan fungsi tertentu yang telah dirancang. Contoh nyata dari robot ini adalah robot manipulator berlengan.

 Mobile robot

Mobile dapat diartikan bergerak, sehingga robot ini dapat memindahkan dirinya dari satu tempat ke tempat lain. Robot ini merupakan robot yang paling populer dalam dunia penelitian robotik. Dari segi manfaat, robot ini diharapkan dapat membantu manusia dalam melakukan otomasi dalam transportasi, platform bergerak untuk robot industri , eksplorasi tanpa awak, dan masih banyak lagi. Robot line follower termasuk kedalam mobile robot.

 Gabungan mobile robot dan non mobile robot

Robot ini merupakan penggabungan dari fungsi-fungsi pada robot mobile dan non-mobile. Sehingga keduanya saling melengkapi dimana robot nonmobile dabat terbantu fungsinya dengan bergerak dari satu tempat ke tempat lain.

 Humanoid

Robot ini sengaja dirancang dengan menirukan manusia. Fungsi-fungsi tubuh manusia baik lengan, kaki, mata, dan pergerakan sendi kepala dan bagian lainnya sebisa mungkin diterapkan dirobot ini. Contoh robot ini adalah robot ASIMO buatan Jepang.

Robot line follower merupakan robot yang termasuk ke dalam mobile robot. Robot ini difungsikan untuk dapat mengikuti garis pada suatu lantai dan kemudian bergerak sesuai dengan pola garis tersebut. Robot ini menggunakan sensor yang memancarkan cahaya yang kemudian ditangkap oleh photodiode, yang apabila terkena garis putih photodiode akan menerima cahaya pantulan dengan maksimal. Sebaliknya jika terkena garis hitam, photodiode akan menerima cahaya yang minimum sehingga dihasilkan tegangan yang kecil.

IC

Integrated Circuit atau IC) adalah komponen dasar yang terdiri dari resistor, transistor dan lain-lain. IC adala

h komponen yang dipakai sebagai otak peralatan elektronika.

Pada komputer, IC yang dipakai adalah mikroprosesor. Dalam sebuah mikroprosesor Intel Pentium 4 dengan ferkuensi 1,8 trilyun getaran per detik terdapat 16 juta transistor, belum termasuk komponen lain. Fabrikasi yang dipakai oleh mikroprosesor adalah 60nm.

Sirkuit terpadu dimungkinkan oleh teknologi pertengahan abad ke-20 dalam fabrikasi alat semikonduktor dan penemuan eksperimen yang menunjukkan bahwa alat semikonduktor dapat melakukan fungsi yang dilakukan oleh tabung vakum. Pengintegrasian transistor kecil yang banyak jumlahnya ke dalam sebuah chip yang kecil merupakan peningkatan yang sangat besar bagi perakitan tube-vakum sebesar-jari. Ukuran IC yang kecil, tepercaya, kecepatan "switch", konsumsi listrik rendah, produksi massal, dan kemudahan dalam menambahkan jumlahnya dengan cepat menyingkirkan tube vakum.

Hanya setengah abad setelah penemuannya, IC telah digunakan dimana-mana. Radio, televisi, komputer, telepon selular, dan peralatan digital lainnya yang merupakan bagian penting dari masyarakat modern. Contohnya, sistem transportasi, internet, dll tergantung dari keberadaan alat ini. Banyak skolar percaya bahwa revolusi digital yang dibawa oleh sirkuit terpadu merupakan salah satu kejadian penting dalam sejarah umat manusia.

IC mempunyai ukuran seukuran tutup pena sampai ukuran ibu jari dan dapat diisi sampai 250 kali dan digunakan pada alat elektronika seperti:

• Handphone
• Radio

Hard to believe

I was sooo happy yesterday because of the fact that I could really get the thing I want next year hope it will come in the early year! wohoooo! so excited! BUUUT my happiness yesterday, it seemed gone through the night. Some of my friends really made me down I hate it! A lot of things I've learned and I will always remember that deep in my memory. helooo guys, it shows that we've almost reached the sahur time!! because it is now 2:58 AM!!
OMO I haven't gone to sleep tonite.. A lil bit tired but I had a LOTTA FUN! I'll tell you the complete story....................

It began at 22:34 pm when I accidentally met my friend --> Sarah Mayangsari on Twitter. Well we've been such a very good friend since the second grade of high school cause we were chairmate!! Hufht I miss her :''''''( but.. I am now with Mira Khairunnisa! She's sooo kind, we've gotten along pretty much so I like her too as much as you Sar! ----back to main topic.

A few days before, sarah and I have planned to msn-ing or skype-ing together to do an assignment. We'll have to do a quiz from the SMAN 47 Cyberschool. Cool isn't it? haha. then We'll see the score from the answers we had submitted. First, after preparing ourselves, then we start the quizzzzzz

It was rather difficult for us to answer the quiz. It's not that easy than I think. We stuck at the last questions something called Peter. Peter Parker(?) OF COURSE NOT!

At that critical time, we were goin' a lil' bit crazy haha. Don't ever think about it, cause this is only between me, Sarah, and our blog LOL. We're chatting through the Msn. We drew some "pictures" *not for children under17* and we also made some abbreviation. For example : BeTi ( B**a on Twitter hahaha ROFL. We laughed all night because of that particular joke!! Then, we finally made a decision. We had chosen the answer.

We were facing several hurdle initially but.. after asking some answers from Mira and checking at the dictionary we finally submitted all the answers! and....... We got 75! I rather dissapointed but I have to admit it, this is our hard work ke ke ke.

Yeah that was a fun night, I hope I could get another crazy night with Sarah.. LOL lupu

Crazy Hour

I cannot sleep well these days. Maybe it's because of the "afternoon-sleeping routine" which made me insomnia. My laptop and the internet became my best friends at these crazy hour. I can't imagine who would accompany me if they're not there. I love you my friend
